Description
Meganerv 300: Vitamin B1 + B6 + B12 (MEGANERV 300 Capsule) is a high potency formula of the three neurotropic vitamins. Thiamine mononitrate (B1), pyridoxine hydrochloride (B6) and cyanocobalamin (B12). This combination of the major B-complex vitamin is specifically valuable in the comprehensive management of various forms of neuropathies.
Meganerv 1000: Vitamins B1 + B6 + B12 (MEGANERV 1000) tablet is a scientifically based high potency combination of the neurotropic vitamins B1 + 86 + B12. This pharmacodynamic and neurotropic formulation derives its dual capacities for etiologic therapy and symptomatic treatment of neuropathic and neuralgic conditions from the complementary operative relationships among its three active constituents.
Vitamins B1 + B6 + B12 (MEGANERV 1000) tablet is coated with a single film integument which masks the unpleasant tastes of its constituted ingredients and gives the tablet a delectable appearance. Moreover, this film coat protects its active components from possible loss of potency caused by light and other atmospheric degradative factors.

Pharmacology: Pharmacodynamics: Normal cellular activities depend on the availability of the biochemical energy derived mainly from the food nutrients. In the metabolic systems of 1he body, the substrates or breakdown products of carbohydrates, fats and proteins must be acted upon by enzymes together with their co-enzymes (primarily vitamins B1, B6 and B12).
B vitamins are necessary in order that the processes by which these substrates are further transformed may continue. Vitamin B1 + B6 + B12 stimulate the various metabolic processes which produce the energy needed to fuel the cellular activities of the body, specifically the physiologic activities of the nervous system. As the well known actions of B1, B6 and B12 are directed towards the CNS, the purported actions of high doses of these major vitamins in Vitamin B1 + B6 + B12 (MEGANERV 300 Capsule) revolve around the central nervous system. Vitamin B1 + B6 + B12 (MEGANERV 300 Capsule) has sufficient concentration of B1, thiamine mononitrate. Thiamine Mononitrate as a co-enzyme of carbohydrate metabolism, ensures the efficient production of energy from glucose into other metabolites such as ribose, a major component of DNA and RNA. Glucose provides the basic nutritional needs of the nervous system. The cells of the nervous system depend entirely on glucose as its energy source. Independent of its co-enzyme function, thiamine acts as a modulator in neuromuscular transmission. Vitamin B1 + B6 + B12 (MEGANERV 300 Capsule), with its high concentration of pyridoxine hydrochloride, is involved in several metabolic transformations of amino acids for tissue building and repair and in the biosynthesis of certain compounds such as neurotransmitters and blood elements needed for hematopoiesis.
Vitamin B1 + B6 + B12 (MEGANERV 300 Capsule) contains substantial amount of cyanocobalamin, a co-enzyme which promotes metabolism of nucleic acids, fats and proteins. Therefore, it is necessary in the formation of new cells and in the repair of nerve tissues.
The high amounts of B1, B6 and B12 in MEGANERV 300 Capsule will help optimize nutrition of the neurons thereby promoting efficient functioning of the nervous system.
Pharmacokinetics: Following oral administration, thiamine is absorbed from the gastrointestinal tract and is widely distributed to most body tissues. Thiamine is mostly present within the cells as diphosphate.
Thiamine is not stored in the body and amounts in excess of the body's requirements are excreted in the urine as unchanged thiamine or as metabolites.
Pyridoxine is readily absorbed from the gastrointestinal tract. It is converted to the active form pyridoxal phosphate and is stored mainly in the liver. Amounts in excess of the body's requirements are excreted unchanged in the urine. Pyridoxal diffuses across the placenta and appears in breast milk.
Vitamin B12 compounds which bind to intrinsic factor, a glycoprotein secreted by the gastric mucosa are then actively absorbed from the gastrointestinal tract. Little amount is absorbed by passive diffusion but the process becomes increasingly important with larger or therapeutic amounts.
Vitamin B12 is extensively bound to transcobalamin, a plasma protein. Vitamin B12 is stored in the liver and is excreted in the bile. Part of a dose is excreted in the urine. Vitamin B12 diffuses across the placenta and is also distributed into breast milk.
Indications/Uses
Meganerv 300: The therapeutic Vitamin B1 + B6 + B12 (MEGANERV 300 Capsule) act in close functional relationship with one another to provide rapid, decisive clinical response in the following disorders and conditions: Painful neurological manifestations such as neuritis, polyneuritis, neuralgia, lumbago, ischialgia, sciatica, cervical and shoulder-arm syndrome. Neuropathies caused by certain disease states such as diabetes and cardiac disorders. Alcoholic neuropathy; iatrogenic complications arising from INH, reserpine and phenothiazine therapy; other drug-induced neuropathies. Hyperemesis gravidarum: neuropathic changes during pregnancy.
Meganerv 1000: Vitamins B1 + B6 + B12 (MEGANERV 1000) tablet is indicated in the following conditions and disorders: Painful neurological manifestations such as neuritis, polyneuritis, lumbago, ischialgia, sciatica, cervical and shoulder-arm syndrome.
Neuropathies caused by certain disease states such as diabetes and cardiac disorders.
Alcoholic Neuropathy: Iatrogenic complications arising from isoniazid, reserpine and phenothiazine therapy; other drug induced neuropathies.
Hyperemesis gravidarum; neuropathic changes during pregnancy.
Dosage/Direction for Use
Meganerv 300: For therapeutic use, 2-4 capsules should be administered daily. Chronic cases may require longer therapy which the physician will have to adjust depending on the needs of the patient.
For prophylactic administration when diseases or drugs are likely to lead to neurological complications, 1-2 capsules daily is recommended.
Meganerv 1000: For therapeutic use, 2-4 tablets of Vitamins B1 + B6 + B12 (MEGANERV 1000) should be administered daily until acute symptoms subside. Chronic cases may require longer therapy.
For prophylactic administration when diseases or drugs are likely to lead to neurological complications, the recommended dosage is 1-2 tablets daily.
Contraindications
Contraindicated in patients with history of hypersensitivity to the components. Cyanocobalamin should not be given to patients with suspected vitamin B12 deficiency without confirmation of the diagnosis.
Special Precautions
Meganerv 300: Cyanocobalamin (Vitamin B12) should not be used to treat megaloblastic anemia of pregnancy. Administration of doses greater than 10 mcg of Vitamin B12 daily may produce a haematological response in patients with folate deficiency; indiscriminate use may mask the precise diagnosis. Cyanocobalamin or hydroxocobalamin should, if possible, not be given to patients with suspected vitamin B12 deficiency without first confirming the diagnosis.
Regular monitoring of the blood is advisable.
Meganerv 1000: The vitamins in Vitamins B1 + B6 + B12 (MEGANERV 1000) are non-toxic even in high doses and usually no adverse or after effects occur. However, sensitivity to Vitamin B1 may sometimes be encountered. Sensitivity may occur if administration at prolonged intervals is irregular, or if the initial dose is low and is suddenly increased.
Liver dysfunction also seems to predispose patients to hypersensitivity reactions. Nevertheless, it must be stressed that hypersensitivity to Vitamin B1 is very rare when given orally and is encountered more often only when given by intravenous injection.
Use In Pregnancy & Lactation
Meganerv 300: Cyanocobalamin (Vitamin B12) should not be used to treat megaloblastic anemia of pregnancy.
Drug Interactions
Drugs that increase the requirements for pyridoxine include hydralazine, isoniazid, penicillamine and oral contraceptives.
Neomycin, aminosalicylic acid, histamine H2-antagonists and colchicine may reduce the absorption of vitamin B12 from the gastrointestinal tract. Oral contraceptives may reduce serum concentrations of vitamin B12. Though unlikely to be of clinical significance, many interactions of vitamin B12 should be taken into account when performing assays for blood concentration. Parenteral chloramphenicol may attenuate the effect of vitamin B12 in anemia.
